Figure: Differential Scanning Fluorimetry Curve. Fluorescence Intensity (arbitrary units) vs Temperature (°C). Observed intensity, black '+'; smoothed observations, black curve; final fitted model, red curve; residual (smoothed observations - fit), green curve; background, blue curve. Transition midpoint temperature (Tm) and transition intensity range from Imin to Imax, vertical thick blue bar(s); transition temperature range (full width at half maximum slope, FWHM) and intensity at transition midpoint (ITm), horizontal thick green bar(s).
Curve Fitting: Unsmoothed observed fluorescence intensity is fit with a background and one or more intensity transitions In:
    I = Ibg + I1 + I2 + … where     In = ΔI / {1 + e(TM - T) / TW } ,   ΔI = Imax - Imin and   TW is proportional to the full width at half maximal slope (FWHM).   Ibg = Imin for the first transition (constant) unless exponentially decaying background is allowed with the -x option and decay is detected, in which case either:
    Ibg = Iinit × ekT (decay from Iinit to zero)
or Ibg = Imin + (Iinit - Imin) × ekT (decay from Iinit to Imin). When Imin is far below zero, this gives a nearly linear decreasing background.
Table: Transition Models. Overall data problems, if any, in text above table. If no transition found, no table is displayed. Problems with fitting, if any, to the right. Pale green background, transition(s) with maximum slope or ΔI. Since these may be in different rows, transition are numbered from low to high Tm rather than by slope or ΔI. Tm, transition midpoint; s.d., deviation of observed from expected intensity near Tm. Note: this s.d. indicates data and fit quality of one experiment, not expected deviation between experiments. Slope, (dI/dT)max = dI/dT at Tm from model. Delta, transition intensity change ΔI relative to maximum observed intensity: ΔI / Imax obs (and relative to the largest ΔI: ΔI / ΔImax). For data baselined to minimum intensity, a common default setting, Imax obs is the total observed intensity change. If that change is largely due to intensity decay, then Σ (ΔIi / Imax obs) << 1.
